O fungo, Aspergillus flavus, prospera em plantas de milho que s\u00e3o estressadas pelo clima quente e seco, diz o professor assistente de pesquisa Ron Sayler. A aflatoxina raramente \u00e9 uma preocupa\u00e7\u00e3o para os produtores de milho no Centro-Oeste por causa do clima mais frio de l\u00e1.<\/p>\n<p>No Arkansas, a irriga\u00e7\u00e3o \u00e9 altamente recomendada para o milho para reduzir o risco de contamina\u00e7\u00e3o por aflatoxina; no entanto, isso ocorre frequentemente mesmo em campos irrigados, particularmente em um ver\u00e3o muito quente e seco, como ocorreu em 2010, disse Sayler. O projeto \u00e9 financiado em parte pelo Arkansas Corn and Sorghum Promotion Board.<\/p>\n<p>Foi comprovado que um gene no feij\u00e3o-jacinto inibe a atividade da enzima alfa amilase do A. flavus necess\u00e1ria tanto para o crescimento f\u00fangico quanto para o ac\u00famulo subsequente de aflatoxinas, disse Sayler. O patologista vegetal Charles Woloshuk isolou o gene h\u00e1 cerca de 10 anos na Purdue University, onde Bluhm era membro da equipe de pesquisa antes de vir para o Arkansas. A inser\u00e7\u00e3o do gene no milho n\u00e3o estava sendo buscada, ent\u00e3o Bluhm e Sayler pegaram o projeto no Arkansas.<\/p>\n<p>Sayler desenvolveu um segmento de DNA contendo o gene e introduziu o transgene na linhagem de milho Hi II.<\/p>\n<p>Sayler relatou que, \u201co membro da fam\u00edlia do gene inibidor de amilase B01 foi clonado no vetor de transforma\u00e7\u00e3o de planta 1300S, que \u00e9 projetado para expressar B01 em todos os tecidos (da planta de milho). Vinte e uma linhagens com o transgene inibidor de amilase B01 foram regeneradas e est\u00e3o sendo cultivadas at\u00e9 a maturidade na estufa. As sementes de cada uma dessas linhagens ser\u00e3o testadas quanto \u00e0 sua capacidade de inibir a infec\u00e7\u00e3o por Aspergillus flavus e o ac\u00famulo de aflatoxina.\u201d<\/p>\n<p>\u201cQuando voc\u00ea introduz um gene no milho, ele vai para diferentes partes do cromossomo do milho. Onde o gene est\u00e1 localizado afeta a express\u00e3o da caracter\u00edstica gen\u00e9tica desejada\u201d, disse Sayler. Testes de sementes de cada linha transg\u00eanica determinar\u00e3o qual linha inibir\u00e1 significativamente o crescimento do fungo e a produ\u00e7\u00e3o de aflatoxina. Os resultados devem ser conhecidos neste ver\u00e3o, disse ele.<\/p>\n<p>\u201cEstou otimista de que uma das linhas expressar\u00e1 n\u00edveis (de inibidor de amilose) altos o suficiente para inibir o crescimento do fungo\u201d, disse Sayler.
